Polymeric delivery systems for nucleic acid therapeutics: Approaching the clinic
نویسندگان
چکیده
Gene therapy using nucleic acids has many clinical applications for the treatment of diseases with a genetic origin as well development innovative vaccine formulations. Since in their free form are rapidly degraded by nucleases present extracellular matrices, have poor pharmacokinetics and hardly pass cellular membranes, carrier systems required. Suitable carriers that protect acid payload against enzymatic attack, prolong circulation time after systemic administration assist binding internalization needed to develop based drug products. Viral vectors been investigated also clinically used delivery vehicles. However, some major drawbacks associated use. Therefore there substantial attention on use non-viral cationic lipids polymers. This review focuses properties polymer-based formulations, referred polyplexes. Different polymeric summarized, barriers polyplexes encounter ways tackle these discussed. Finally is given status

Nucleic acid therapeutics
The discovery of ribozymes by Cech and Altman has fundamentally changed the view of the function of RNA in chemistry, biology, and medicine (1–3). Prior to this discovery, RNA had been viewed as a passive molecule that only carried information or provided structure to RNA-protein complexes. It is now clear that RNA can act as an enzyme and is capable of catalyzing RNA splicing and cleavage, as ...

As alterations in gene expression have become a better understood component of normal development and disease pathogenesis, transcription factors and other regulators of gene expression have become an increasingly attractive target for potential therapeutic intervention. Transcription factors are generally nuclear proteins that play a critical role in gene regulation and can exert either a posi...

Atherosclerosis arises from an excessive fibroproliferative-inflammatory response to injury of the artery wall and typically evolves over decades (1). Smooth muscle cells play a central role in the development of atherosclerotic lesions. Their ability to migrate from the medial compartment of the artery wall to the intima, where they proliferate, accumulate lipid, and generate extracellular mat...
متن کاملFunctionalized CNTs for delivery of therapeutics
Carbon nanotubes (CNTs) are high aspect ratio allotropes of carbon. Because of their unique physical and chemical characteristics, they are nominated for a vast variety of applications in the biomedical field. Their very low solubility is the only problem of them which is solved by different methods of Functionalization. After discoveries in the last few years of the capacity of CNTs to penetra...
متن کاملNano-vectors for the Ocular Delivery of Nucleic Acid-based Therapeutics
Nucleic acid-based therapeutics have gained a lot of interest for the treatment of diverse ophthalmic pathologies. The first to enter in clinic has been an oligonucleotide, Vitravene(®) for the treatment of cytomegalovirus infection. More recently, research on aptamers for the treatment of age related macular degeneration has led to the development of Macugen(®). Despite intense potential, effe...
